流式细胞术检测水中细菌总数的方法研究

摘要:

环境水检测中最常见的检测为细菌计数。常规的计数方法主要是平板计数法和通用的显微计数法,但是这两种方法误差大、耗时长,且平板法只能计数活菌。流式细胞术用于菌体计数可以快速得到细菌总数,若检测的是一定体积样品中的菌数,即可得知菌浓度,同时可区分活菌和死菌,获得活菌百分比。研究了流式细胞术检测环境水中细菌总数的方法,采用荧光性染料进行染色,不同稀释度水样进入流式细胞仪分析、计数。同时与国家标准检测方法进行比较,验证了其准确性和灵敏度。结果显示,在一定范围浓度下,流式细胞术检测环境水中的细菌总数与平板法成线性相关,检测结果的常用对数值基本一致,有很好的符合性。利用SYBR Green和DAPI可以检测活菌和死亡的菌体,更能反应出实际细菌总数。流式细胞术是一种比平板法检测细菌总数更加快速、准确的方法,极大地缩短了检测时间。

Abstract:

The most common detection in environmental water detection is bacterial count. The conventional counting methods are mainly plate culture and general microscopy,but these two methods have large error and long time-consuming,and the plate method can only count live bacteria. The total number of bacteria can be quickly obtained by flow cytometry for cell counting. If the number of bacteria in a certain volume of sample is detected,the bacterial concentration can be obtained. At the same time,live bacteria and dead bacteria can be distinguished to obtain the percentage of live bacteria. The method of detecting the total number of bacteria in environmental water by flow cytometry was studied. Fluorescent dyes were used for dyeing,and water samples with different dilutions were analyzed and counted by flow cytometry. At the same time,compared with the national standard detection method,its accuracy and sensitivity are verified. The results showed that there was a linear correlation between the total number of bacteria in the environmental water detected by flow cytometry and the plate method,the common logarithm values of the test results were basically consistent and had good consistency. SYBR green and DAPI can be used to detect living bacteria and dead bacteria,which can better reflect the actual total number of bacteria. Flow cytometry is a faster and more accurate method to detect the total number of bacteria than plate method,which greatly shortens the detection time.
