硫酸盐还原菌的生长因子分析及脱硫性能研究

Growth factor analysis and desulfurization performance of sulfate-reducing bacteria

摘要:

硫酸盐还原菌(SRB)是一类利用有机物还原SO4 2-产生硫化物的细菌。SRB可用于处理含SO4 2-废水,但是脱硫效果受温度、环境pH、S2-质量浓度、m(COD)/m(SO4 2-)等因素限制。基于此,本研究采用批量实验,综合分析温度、环境pH、S2-质量浓度和m(COD)/m(SO4 2-)这4个影响因素对SRB生长的影响,并探究SRB在不同环境下的脱硫性能。结果表明:培养14~86 h时,SRB处于对数期,此时其活性最高;SRB的最佳生长温度为35 ℃;体系中的S2-会影响SRB生长,当S2-质量浓度增加时,不仅会抑制SRB的代谢活性,甚至可导致SRB细胞凋亡;SRB能在环境pH为5~8的条件下存活,当pH为7~8时,SRB的代谢最旺盛;最适宜SRB生长的m(COD)/m(SO4 2-)为2。在最佳条件,即温度35 ℃、pH=7、m(COD)/m(SO4 2-)=2条件下,采用Starkey培养基培养处于对数期的SRB 8 d后,溶液OD600、pH、ORP、电导率(EC)、SO4 2-去除率分别为1.33、8.78、-393 mV、2.90 mS/cm、84.77%。

Abstract:

Sulfate-reducing bacteria (SRB) was a class of bacteria that use organic matter to reduce SO4 2- to produce sulfides. SRB can be used to treat wastewater containing SO4 2-,but the desulfurization effect is limited by temperature,environmental pH,S2- concentration,m(COD)/m(SO4 2-) and other factors. Based on this,this study used batch experiments to comprehensively analyze the effects of temperature,environmental pH,S2- mass concentration and m(COD)/m(SO4 2-) on the growth of SRB,and explored the desulfurization performance of SRB in different environments. The results showed that SRB was in the logarithmic phase when cultured for 14-86 h,and the activity was the highest. The optimum growth temperature of SRB was 35 ℃. The S2- in the system would affect the growth of SRB. When the mass concentration of S2- increased,it would not only inhibit the metabolic activity of SRB,but also lead to apoptosis of SRB cells. SRB could survive under the condition of pH 5-8. When the pH of the environment was 7-8,the metabolism of SRB was the most vigorous. The optimum m(COD)/m(SO4 2-) for SRB growth was 2. Under the optimal conditions of temperature 35 ℃,pH=7,and m(COD)/m(SO4 2-)=2,SRB in logarithmic phase was cultured on Starkey medium for 8 days,the OD600,pH,ORP,EC,and SO4 2- removal rates of the solution were 1.33,8.78,-393 mV,2.90 mS/cm,and 84.77%,respectively.
